Our research is focused on resolving complex structures and dynamic processes of soft colloids using the unique ability of cryo electron microscopy methods, cryo-EM, to simultaneously reveal coexisting structures, capture short-lived intermediates, and directly illuminate structural details. This information, importantly, relates to the native, bulk state, and is at nanometer resolution. Here we highlight traditional and new directions in Cryo-EM that combined provide clearer and more precise structural, spatial, temporal and quantitative data of colloidal systems, and much improved understanding of structure, properties and function.
The traditional and leading technique in this family, Cryo-TEM, is powerful as it directly and without image processing discloses incredible morphologies and structures that are many times beyond what we could anticipate or imagine. Until recently however, we could only collect single images, 2D projections of the sample volume, and the spatial organization remained an unexplored domain. With cutting edge technological improvements in detection by direct detectors and in optics through phase plates, and by advancing new methodologies including cryo-electron tomography (Cryo-ET), we can now address questions in colloid science of additional complexity and unfold the 3D organization of soft nanosized colloids in solution. Examples discussed will include micellar systems, delivery vehicles, 1-D structures and more.
